Background:
An oxidoreductase that catalyzes the reaction between superoxide anions and hydrogen to yield molecular oxygen and hydrogen peroxide. The enzyme protects the cell against dangerous levels of superoxide.
Description :
Recombinant Human Cu/Zn Superoxide Dismutase produced
in E.Coli is a homodimer, non-glycosylated polypeptide chain
containing 2 x 154 amino acids and having a total molecular mass
of 31,608 Dalton.
The Cu/Zn SOD is purified by proprietary chromatographic
techniques.
Physical Appearance:
Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation:
Recombinant Superoxide Dismutase was lyophilized after from a sterile solution containing 50mM sodium phosphate
buffer and 100mM sodium chloride.
Solubility:
It is recommended to reconstitute the lyophilized SOD in sterile 18M-cm H2O not less than
100ug/ml, which can then be further diluted to other aqueous solutions.
Stability:
Lyophilized Superoxide Dismutase although stable at room temperature for 2 weeks,
should be stored desiccated below -18 C. Upon reconstitution should be stored at 4 C.

Purity:
Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino acid sequence:
The sequence of the first five N-terminal amino acids was determined and was found to
be Ala-Thr-Lys-Ala-Val.
Dimers and aggregates:
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Biological Activity:
ProSpec's recombinant Superoxide Dismutase is fully biologically active when compared to standard.
The potency per mg was tested by Pyrogallic Acid method and was found to be 8232Units/mg.
Endotoxin:
Less than 0.1 ng/ug (IEU/ug) of SOD.
Protein content:
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a standard solution of SOD as a Reference Standard.
