Background:

Mitogen-activated protein kinases (MAPKs) have been implicated in the signal transduction of the endothelial response to growth factors and inflammatory stimuli. MEK1, a member of the mitogen-activated protein kinase (MAPK) cascade directly activates extracellular signal-regulated kinase (ERK). MEK1 and MEK2 are closely related, dual-specificity tyrosine/threonine protein kinases found in the Ras/Raf/MEK/ERK mitogen-activated protein kinase (MAPK) signaling pathway. Approximately 30% of all human cancers have a constitutively activated MAPK pathway, and constitutive activation of MEK1 results in cellular transformation.

Description :

Recombinant Human MEK1 produced in Sf9 insect cells is a glycosylated, polypeptide chain containing amino acids 2-393 and having a molecular mass of 47 kDa. Recombinant MEK1 is purified by proprietary chromatographic techniques. This protein is the full-length form of the protein with a poly His-tag.

Formulation:

Recombinant Human MEK1 is supplied at a concentration of 0.23mg/ml in 20mM Tris, pH 8.0, 0.15M NaCl, 0.27M sucrose, 1mM DTT, 1mM benzamidine, 0.2mM PMSF and 0.03% Brij-35.

Biological Activity:

Recombinant human MEK1 is phosphorylatable in vitro, using either recombinant active MEKK1, MEKK1 immunoprecipitated from stimulated cells, c-Raf immunoprecipitated from stimulated cells, or stimulated cell extracts. This phosphorylation can be monitored by Western blot analysis using phosphorylation site specific antibody directed to MEK1/2 [pS222], in conjunction with chemiluminescence detection methods.

Storage:

Store at 4蚓 if entire vial will be used within 1-2 weeks. Store frozen at -20蚓 for longer periods of time.
Please avoid freeze-thaw cycles.

Purity:

Greater than 95% (SDS-PAGE).
