Background:
Placental growth hormone, or growth hormone variant (GH-V), is a member of the growth hormone gene family. Its physiologic role during pregnancy remains undefined. Although extensive work has been done characterizing the signaling pathways activated by hGH, the signaling pathways which are activated by GH-V have not been characterized.
Human placental GH (hPGH) replaces pituitary GH during pregnancy. hPGH is correlated to serum IGF-I in normal pregnancies and in pregnancies complicated by fetoplacental disorders
Description:
Recombinant Placental Human GH-22K produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 192 amino acids and having a molecular mass of 22367 Dalton. Predicted pI=7.80.
Recombinant Human GH-22K-pl and Recombinant Human GH-22K-pit have similar biological activity mediated through human growth hormone receptors. However Recombinant GH-22K-pl is completely devoid of any lactogenic (prolactin receptor mediated) activity in homologous system consisting of human prolactin receptors, an activity which is characteristic of human pituitary 22 K and 20 K GHs. Thus, in contrast to pituitary hGHs it can be preferably used in any system in which the accompanying lactogenic activity is not wanted.
Recombinant Human GH-22K-pl is purified by proprietary chromatographic techniques.
Physical Appearance:
Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation:
The protein was lyophilized from a concentrated (1mg/ml) solution with 0.0045mM NaHCO3 previously adjusted pH 8-9.
Solubility:
It is recommended to reconstitute the lyophilized Recombinant GH-22K-pl in 0.4% NaHCO3 or water adjusted to pH 9, not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Stability:
Lyophilized Human GH-22K-pl although stable at room temperature for 3 weeks, should be stored desiccated below -18
0C. Upon reconstitution and filter sterilization Recombinant GH-22K-pl can be stored at 4
0C for up to 4 weeks.
For long term storage and more diluted solutions it is recommended to add a carrier protein (0.1% HSA or BSA).
Please avoid freeze-thaw cycles.
Purity:
Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Gel filtration at non denaturing conditions using 25 nM Tris-HCl + 150 nM NaCl, pH 8.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained.
Amino acid sequence:
The sequence of the first five N-terminal amino acids was determined and was found to be
Ala-Phe-Pro-Thr-Ile.
Dimers and aggregates:
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Biological Activity:
ProSpec旧 Recombinant Human Placental Growth Hormone-22K is fully biologically active when compared to World Health Organization (WHO) reference standard.
Endotoxin:
Less than 0.1 ng/ug (IEU/ug) of Recombinant Placental Growth Hormone.
Protein content:
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 1.18 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a calibrated solution of GH-22K-pl as a Reference Standard.
