Background:

Follistatin (FS), a glycoprotein, plays an important role in cell growth and differentiation through the neutralization of the biological activities of activins. Follistatin (FST) is a monomeric activin-binding and neutralization protein that has at least three isoforms in human tissues and fluids. The full-length FS315 protein has an acidic 26-residue C-terminal tail that is not present in the shortest form, FS288, due to alternative splicing. An intermediate form, FS303, was identified in follicular fluid that is presumably derived by proteolytic processing of this tail domain. Interestingly, the biochemistry of each of these three isoforms is distinct, including their ability to bind to cell surface proteoglycans, an activity that ranks in the order FS288 > FS303 > FS315.

Description :

Recombinant Human Follistatin produced in E.Coli is a single, non-glycosylated polypeptide chain containing 288 amino acids and having a total molecular mass of 31.5kDa. Recombinant FST is purified by proprietary chromatographic techniques

Physical Appearance:

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation:

Lyophilized from a concentrated (1mg/ml) solution containing no additives

Solubility:

It is recommended to reconstitute the lyophilized rHuFST in sterile 18MΩ-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.

Stability:

Lyophilized Follistatin although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution FST should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please avoid freeze-thaw cycles.

Purity:

Greater than 95.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Amino acid sequence:

The sequence of the first five N-terminal amino acids was determined and was found to be
Gly-Asn-Cys-Trp-Leu.

Dimers and aggregates:

Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Biological Activity:

ProSpec旧 Recombinant Follistatin is fully biologically active when compared to standard.
The activity is determined by the ability to neutralize Activin A inhibitory effect of mouse MPC-11 cells. The expected ED50 is 0.1-0.4 ug/ml in the presence of 7.5 ng/ml Activin A.

Endotoxin:

Less than 0.1 ng/ug (IEU/ug) of Follistatin.

Protein content:

Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a standard solution of Myostatin as a Reference Standard.
