Background :
Focal adhesion kinase (FAK) is a critical mediator of signal transduction by integrins and growth factor receptors in a variety of cells including endothelial cells (ECs).It is a nonreceptor protein tyrosine kinase involved in signaling downstream of integrins, linking bacterial detection, cell entry, and initiation of proinflammatory response through MAPKs and NF-kappaB activation.
Description :
Recombinant human FAK (amino acids 2 to 1052) is produced in Sf9 cells with an amino-terminal polyhistidine tag and purified via sequential chromatography.
Formulation:
rHuFAKis supplied at a concentration of 0.1mg/ml in 40mM Tris, pH 7.5, 0.15M NaCl, 0.27M sucrose, 1mM DTT, 0.1mM EGTA, 0.2mM PMSF, 1mM benzamidine, 0.1mM sodium vanadate, and
0.03% Brij-35.
Storage:
Store at 4蚓 if entire vial will be used within 1-2 weeks.
Store, frozen at -20蚓 for longer periods of time.
Please avoid freeze-thaw cycles.
Purity:
Greater than 95% (SDS-PAGE).
Biological Activity:
Recombinant Human FAK contains14units/mg. One unit of FAK activity transfers 1 nmole of phosphate to
poly [GluTyr] substrate per minute at 30蚓 in a reaction mixture containing 8mM ATP. Substrates for FAK include STAT, paxillin, p85 PI 3-kinase, Cas, and tensin.FAK also undergoes autophosphorylation.
