Background:

PPK catalyzes the reversible transfer of phosphate between polyphosphate and ATP The phosphorylation of ADP to ATP by polyphosphate kinase is by a processive mechanism; the phosphorylation occurs without release of the polymer from the enzyme prior to termination of the reaction.

Description :

Polyphosphate Kinase purified circa 10 fold . Free of all Polyphosphate Glucokinase activity. Native MW = 83 kD (monomer)

Formulation:

Contains polyphosphates and phosphate buffer salts.

Stability:

One year when stored at -20蚓, three weeks at room temperature.
Please avoid freeze-thaw cycles.

Required cofactors:

The reaction requires Mg2+.

Unit definition:

1 Unit of activity = the amount of PPK required to convert 1 micromole ADP to ATP per minute at 30C and pH 7.4, in phosphate buffer, using polyphosphate as phosphate donor. (The reaction is reversible.)

Biological Activity:

1-3 U per mg protein

Applications:

The PPK reaction provides an inexpensive means of recycling ATP consumed in enzymatic phosphorylation reactions. This has potential for use in enzymatic oligosaccharide synthesis, which requires phosphorylated sugar intermediates.
