:
Botulinum Toxin Type-A blocks neuromuscular transmission by binding to acceptor sites on motor nerve terminals, entering the nerve terminals and inhibiting the release of acetylcholine. The inhibition occurs as the neurotoxin cleaves SNAP-25, a protein integral to the successful docking and release of acetylcholine from vesicles situated within nerve endings. When injected intramusculary at therapeutic doses it produces partial chemical denervation of the muscle resulting in a localized reduction in muscle activity. In addition, the muscle may atrophy, axonal sprouting may accur, and acetylcholine receptors may develop. There is an evedence that reinnervation of the muscle pay occur, thus slowly reversing muscle denervation produced by the BTX-A.
Botulinum toxin type A is the highly purified protein consiting of a 50 kDa light chain and a 100 kDa heavy chain linked by disulfide bonding and is produced from fermentation of Hall Starin Clostridium Botulinum type A grown in a medium containing casein hydrolysate, glucose and yeast extract. It is purified from the culture solution by dialysis and a series of acid precipitations to a complex consisting of the neurotoxin and several accessory proteins.

Physical Appearance:
Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation:
Each 100IU contain 0.5mg HAS, 30mg dextran, 30mg mannitol and 0.9mg NaCl.
Stability:
Lyophilized Botulinum Toxin Type-A although stable at room temperature for 3 weeks, should be stored desiccated below -18
0C. Upon reconstitution Botulinum Toxin Type-A should be stored at 4
0C between 2-7 days and for future use below -18
0C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.
Purity:
Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE .
Endotoxin:
Less than 0.1 ng/ug (IEU/ug) of BTX-A.
Unit Definition:
1 Unit of BTX-A corresponds to 1 LD50 calculated median intraperitoneal dose in mice.
Specific Activity:
20 Units per nanogram of neurotoxin protein complex.
