Ribonuclease A (RNase A) is an endoribonuclease, that specifically cleaves single-stranded RNA 3' to pyrimidine residues (cytosine, uracil). Thereby, it generates pyrimidine-3'-phosphate or oligonucleotides with terminal pyrimidine-3'-phosphates.

A major application for RNAse A is the removal of RNA from preparations of plasmid DNA. For this application, DNAse free RNAse A is used at a final concentration of 10-100 ug/ml.
Stock conc.: 10mg/mL in ddH₂O
Working conc.: 10-100μg/mL (removal of RNA from plasmid preparations; 1 hr, RT) or 100ng-1ug/mL (preparation of "blunt ends" of double-stranded cDNA).
Caution: Boiling of stock solutions to inactivate residual DNase is not necessary or recommended.
