Ethidium bromide is the most commonly used dye for staining DNA during or after polycrylamide or agarose gel electrophoresis.

Staining Solution: Aqueous solutin 0.25~0.5 ug ethidium bromide/mL. Add 10 uL of the Ethidium Bromide Solution (Stock Solution) to 200mL water (dilution, 1/20000, ethidium bomide 0.5 ug/mL). If you feel the ethidium bomide concentration is too strong, dilute the staining solution further 1:1 and find out the staining solution best suited to your system.

Detection: Illuminate with 254 nm, which will be absorbed by the DNA, in turn exciting the ethidium bromide, thus making visile only the DNA containing sites. At 366 nm, all the ethidium bromide will fluoresce.

Caution: Ethidium Bromide is a powerful mutagen and is moderately toxic. Gloves should be worn when working with solutions that contain this dye, and a mask should be worn when weighing ethidium bromide.
