Description
The NBT (nitro-blue tetrazolium chloride) and BCIP (5-bromo-4-chloro-3'-indolyphosphate p-toluidine salt) is an ideal insoluble substrate for use with alkaline phosphatase. The combination of NBT and BCIP yields an intense, insoluble blue - purple precipitate when reacted with alkaline phosphatase, a popular enzyme conjugate for antibody probes. The intense color can be observed visually, is very stable, and will not fade upon exposure to light.
BCIP/NBT Alkaline Phosphatase Color Development Kit offers a convenient, ready-to-use substrate solution for visualizing alkaline phosphatase activity in immunoblotting and immunohistology.

Application
BCIP/NBT Alkaline Phosphatase Color Development Kit has been developed for use in immunochemistry as an insoluble substrate for the detection of alkaline phosphatase. Common uses are in Western blotting or Dot blotting, and to a lesser extent in Immunohistology.
In WB :
BCIP is the AP-substrate which reacts further after the dephosphorylation to give a blue - purple indigo-dye as an oxidation product. NBT serves herein as the oxidant and gives also a blue - purple dye. It intensifies thereby the color and makes the detection more sensitive.

Note:
If precipitates occur in the color substrate solution, they can be removed by warming up the solution to 50°C.
If the precipitates do not resolve centrifuge the tube prior to use because the precipitates might cause background ; centrifugation will not reduce the overall sensitivity.

Blue Taq differs from regular Taq in that it contains blue dye added to the enzyme which is used to first, identify which reactions have already had Taq Polymerase added; second, act as loading dye. When loading for agarose gel analysis post PCR amplification, no loading buffer is required.

In PCR amplification, please use 2.5U Blue Taq and 10ng-500ng template DNA per 50uL PCR reaction, other parameters according to the main technical parameters of Taq. Mixing is requested before the reaction.
